Cephalexin disrupts the synthesis of the peptidoglycan layer of bacterial cell walls which is responsible for cell wall structural integrity. Peptidoglycan synthesis is facilitated by transpeptidases known as penicillin-binding proteins (PBPs). PBPs bind to the D-Ala-D-Ala at the end of muropeptides (peptidoglycan precursors) to crosslink the peptidoglycan. Cephalexin antibiotics mimic the D-Ala-D-Ala site, thereby competitively inhibiting PBP crosslinking of peptidoglycan.

Application

Cephalexin is a cephalosporin antibiotic used to study the effect of expression, binding, and inhibition of PBP3 and other penicillin-binding proteins (PBPs) on bacterial cell wall mucopeptide synthesis.

سفالکسین سنتز لایه پپتیدوگلیکان دیواره سلولی باکتری‌ها را که مسئول یکپارچگی ساختاری دیواره سلولی است، مختل می‌کند.سنتز پپتیدوگلیکان توسط ترانس‌پپتیدازهایی که به عنوان پروتئین‌های متصل‌شونده به پنی‌سیلین (PBP) شناخته می‌شوند، تسهیل می‌شود.PBPها به D-Ala-D-Ala در انتهای موروپپتیدها (پیش‌سازهای پپتیدوگلیکان) متصل می‌شوند تا پپتیدوگلیکان را به صورت متقاطع پیوند دهند.آنتی‌بیوتیک‌های سفالکسین جایگاه D-Ala-D-Ala را تقلید می‌کنند و در نتیجه به طور رقابتی اتصال عرضی PBP پپتیدوگلیکان را مهار می‌کنند.

کاربرد

سفالکسین یک آنتی‌بیوتیک سفالوسپورینی است که برای مطالعه تأثیر بیان، اتصال و مهار PBP3 و سایر پروتئین‌های متصل‌شونده به پنی‌سیلین (PBPs) بر سنتز موکوپپتید دیواره سلولی باکتری‌ها استفاده می‌شود.
