Description
Ponceau S is the sodium salt of a diazo dye. Ponceau S solution is a suitable reagent for use in electrophoresis studies. The Ponceau S stain comes ready-to-use and is designed for rapid (5 min) staining of protein bands on nitrocellulose or PVDF membranes (Western blots) and also for staining protein on cellulose acetate membranes. It may be employed as stain for the purified fibrinolytic enzyme (CK) for the analysis of N-terminal amino acid sequence of the first 14 residues of CK by a protein sequencer. Ponceau S staining on nitrocellulose and PVDF is reversible by washing the stained membrane with 0.1M NaOH for 1 minute. The stained and destained membrane can then be immunologically detected (Western blotting). A product information sheet is supplied which includes the staining procedure for cellulose acetate, nitrocellulose, and PVDF.

پونسو S نمک سدیم یک رنگ دی‌آزو است. محلول پونسو S یک معرف مناسب برای استفاده در مطالعات الکتروفورز است. رنگ پونسو S آماده استفاده است و برای رنگ‌آمیزی سریع (5 دقیقه‌ای) باندهای پروتئینی روی غشاهای نیتروسلولز یا PVDF (وسترن بلات) و همچنین برای رنگ‌آمیزی پروتئین روی غشاهای سلولز استات طراحی شده است. این رنگ می‌تواند به عنوان رنگ برای آنزیم فیبرینولیتیک خالص شده (CK) برای تجزیه و تحلیل توالی اسید آمینه N-ترمینال 14 باقیمانده اول CK توسط یک توالی‌یاب پروتئینی استفاده شود. رنگ‌آمیزی پونسو S روی نیتروسلولز و PVDF با شستن غشای رنگ‌آمیزی شده با 0.1 مولار NaOH به مدت 1 دقیقه برگشت‌پذیر است. سپس غشای رنگ‌آمیزی شده و رنگ‌زدایی شده را می‌توان به صورت ایمونولوژیکی تشخیص داد (وسترن بلات).
